Purification / Growth Conditions / Treatment: O/N cultures of E. coli donor NR1769 and recipient NR1769/pKM101 inoculated into fresh LB without antibiotics and incubated for 2.5 hours at 37 degrees and 200 rpm shaking. Glow discharged Au finder grids placed carbon side up on LB 2% agar plate. E. coli donor and recipient cultures pipetted onto grid in 1:1 ratio and placed in sealed chamber with a damp paper towel. Plastic container placed inside 37C stationary incubator for 30 minutes.
