Purification / Growth Conditions / Treatment: Cells were grown in Tryptic Soy Broth (TSB) liquid culture overnight with no supplements. The overnight culture was then used to inoculate fresh TSB, grown until an OD of ~2, and plunge-frozen.

Sample Preparation: 4 grids were frozen. Vitrobot (Mark IV) settings: 95% humidity, 2µl sample, Blot force 6, blot time 4 sec. Wait time 0 sec. Drain time 1 sec. Ethane/propane mix. Grid: Quantifoil Cu 2/2 200 mesh. Added 1ul of gold to 4 ul of cells prior to freezing.
